If you are starting/ending in Las Vegas, you should definitely count two days: one day for Route 66 and Grand Canyon and the second day for Antelope Canyon, Horseshoe Bend, and the drive back to Vegas.
The most popular place to visit at the Grand Canyon (and the one we recommend for this itinerary) is the South Rim of the Grand Canyon. You will have a few hours here to explore, recommended stops are Moran Point, Mather Point, and the Rim View Trail.
